Synthesis of gel-in-liposome and hydrogel nanoparticles based on microfluidic vortex focusing for hydrogel-based drug delivery system

Hydrogel has been widely adapted in biomedicines, drug delivery system, and tissue engineering owing to its biocompatibility, facile chemical tunability, and high water retention capacity. However, traditional hydrogels often suffer from insufficient mechanical stability, slow response time to external stimuli, and limited controllability of its drug release dynamics. Miniaturization of bulk hydrogel system into nanoscale has shown promising potential to overcome these limitations, however, the controlled synthesis of hydrogel in nanoparticle forms remain very challenging. In this study, we adapt microfluidic liposome synthesis technique to formulate gel-in-liposome in 100 nm scale and induced photopolymerization to form hydrogel nanoparticles. A set of hydrogel precursor solutions were examined for various types of photoinitiators, precursor composition, and use of additives. Size-controlled synthesis of nanoparticles was enabled using a microfluidic vortex focusing platform, realized by masked stereolithography.
