Injectable and Dual-crosslinkable Biohybrid Thermogel for Tissue Engineering Applications

Thermogels, which can be delivered through minimally invasive methods, have received considerable attention in the biomedical field, but often show low mechanical strength and limited cell adhesion. In this study, a novel biohybrid thermogel with adjustable mechanical properties and improved cell adhesion was prepared. Methacrylic glycol chitosan (MGC) was synthesized through N-methacrylation of glycol chitosan, a biodegradable natural polymer. Dual-crosslinkable biohybrid thermogel was developed through simple mixing with gelatin methacryloyl (GelMA) which provides good cell affinity. MGC/GelMA undergoes gelation at body temperature, and its mechanical properties can be improved through UV crosslinking without cell toxicity. We evaluated physicochemical properties and confirmed improved cell adhesion and high biocompatibility. As a result, MGC/GM hybrid thermogels show great potential as an effective injectable scaffold with customized properties and high biocompatibility.
