Loading and Releasing Local Anesthesia from GelMA beads for Embolization Therapy

Polymeric hydrogels are fascinating platforms as scaffolds for drug delivery system (DDS). Among these scaffolds, Gelatin Methacryloyl (GelMA) has emerged as one of the most promising options for DDS due to its biocompatibility and tunable physicochemical properties, which enable controlled drug release kinetics and excellent bioactive properties, such as enzymatic degradation. Utilizing Local Anesthesia (LA) is a promising approach to alleviate postoperative pain. Unfortunately, LAs are often associated with short half-lives. To overcome these limitations, we investigated novel hydrogel beads via the water-in-oil polymerization technique with different functional monomers containing GelMA to entrap the LA for sustained release. The results show the loading and releasing behavior at various pH values for different monomer formulations of beads, which can be utilized as a drug carrier for embolization therapeutic agents as well as for other postoperative pain management.
