Highly Robust Multilayered Collagen-lipid Nanovesicles for All-trans-retinol stabilization and Efficient Skin Delivery

Lipid-based nanocarriers are frequently used in cosmetics to solubilize and deliver water-insoluble active compounds to the skin effectively. However, structural instability might prematurely release and degrade active chemicals, limiting their practical use. Here, we introduce highly robust multilamellar lipid vesicles (MCLVs) generated via self-assembly of preformed, large unilamellar vesicles (LUVs) with hydrolyzed collagen peptides. MCLVs incorporating All-trans-retinol (ATRL) improve stability and encourage effective skin distribution, withstanding repeated freeze-thaw cycles and surfactants. MCLVs achieve an impressive 97.5% loading efficiency of ATRL and maintain their structural integrity over three months. MCLVs exhibit notably lower cytotoxicity to human dermal fibroblasts. Additionally, collagen expression is effectively upregulated, and MMP is suppressed by the MCLVs. These results suggest the potential of MCLVs as a promising platform for efficient skin delivery of ATRL.
