Localized fat reduction is becoming increasingly important for aesthetic improvement, but traditional liposuction often comes with significant side effects, highlighting the need for safer alternatives. We propose a novel method using poly(ethylene glycol)-based micelles that generate carbon dioxide gas through the hydrolysis of carbonate bonds to induce adipocytolysis. Structural modifications of the micelles enhanced carbon dioxide generation, and the addition of nona-arginine peptides improved their cell-penetrating ability and adipocytolytic effects in vitro. Furthermore, in vivo studies demonstrated substantial local fat reduction without notable side effects. Our findings suggest that these gas-generating micelles may represent a promising strategy for localized fat reduction, with potential applications in aesthetic and biomedical fields.
