3D printed complex tissue structures using self-healable HA-based hydrogels

Hyaluronic acid (HA) is a natural polysaccharide and a major component of the extracellular matrix in tissues. HA-based hydrogels have shown potential in extrusion-based 3D printing to engineer tissues such as bone and cartilage. However, their limited mechanical properties have posed challenges in tissue engineering applications. In this study, we enhanced the toughness and stiffness of HA-based hydrogels while maintaining their self-healing capabilities by synthesizing a diol-modified HA, which was subsequently oxidized to obtain aldehyde-containing HA and forming a double network in the presence of hydrazide-modified HA and sodium alginate. The self-healing properties allowed for extrusion-based 3D printing of the hydrogels without requiring additional post-printing processes. Additionally, we utilized these hydrogels to fabricate a growth plate-mimicking multilayered structure in vivo. Our findings may highlight the potential of these HA-based hydrogels for regenerating complex tissues and organs.
