Polydimethylsiloxane membrane with enhanced adhesion and detachment for cell sheet fabrication

Polydimethylsiloxane (PDMS) membranes can precisely control the generation of well-defined micropores. A large number of microbubbles were generated by mixing the PDMS solution with a Rushton impeller. The microbubbles generated by the impeller generated a large number of small to large micropores in the PDMS membrane. The morphology of the thinner porous PDMS membrane induced higher values for roughness, Young's modulus, contact angle, and air permeability. Keratinocytes were cultured on these membranes, and the cell viability was the highest on the porous PDMS membrane fabricated at a spin-coating speed of 5000 rpm, especially on day 7, due to the internal networking structure and surface properties. Intact detachment of cell sheets attached to porous PDMS membranes was demonstrated. Therefore, PDMS has great potential to enhance cell sheet formation in regenerative medicine.
