Nanostructured Dendritic Cells as a Platform for Enhanced Cancer Immunotherapy

Recent advances in cancer immunotherapy highlight the use of nanotechnology, particularly nanoparticle (NP)-based methods, for enhanced clinical potential in solid cancers. NPs offer a stable platform for personalized therapy by integrating cellular membranes or extracellular vesicles. Dendritic cells (DCs) are key in T cell activation but face challenges like poor in vivo longevity and storage issues. To address this, we developed immortalized nanodendritic cells (anDCs) by coating DC membranes onto gold nanoparticles. These anDCs are thermodynamically stable and retain functionality even after 30 days, showing significant tumor regression and enhanced T cell activation in vivo. By incorporating immune-checkpoint inhibitors like αCTLA-4, anDCs demonstrated strong efficacy in multiple tumor models. This innovation offers a promising approach for personalized cancer treatment, enabling continuous activation of tumor-specific T cells and substantial tumor regression.
