Innovative multimeric genomic hydrogel for efficient and reusable cell-free protein synthesis via dual plasmid multi-primed chain reaction.

Cell-free protein synthesis (CFPS) offers distinct advantages over traditional cell-based protein synthesis methods, including the ability to conduct transcription and translation in vitro. In this study, we present a novel approach for CFPS using a multimeric genomic DNA hydrogel (mGD-gel) synthesized via rolling circle chain amplification (RCCA) with dual single-stranded circular plasmids and multiple primers. This mGD-gel significantly enhances protein yield and can be reused at least five times without losing its structural integrity or protein production capabilities. The mGD-gel’s shape is also customizable, making it adaptable for various applications. This innovative platform demonstrates potential for diverse biotechnological uses, providing a new, efficient, and sustainable method for protein synthesis.
