Overcoming barriers to cell encapsulation for the treatment of diabetes

Pancreatic islet transplantation is a promising treatment for type 1 diabetes because the cells themselves sense glucose and secrete insulin. However, immune system issues necessitate new technologies, such as encapsulation. These immunoprotective polymer capsules allow the selective exchange of gases, nutrients, and small proteins like insulin. They also prevent immune mediator infiltration to reduce immunorecognition. However, translating islet encapsulation technologies to patients involves many challenges. This talk will discuss different encapsulation designs and optimizations. It will address issues like material biocompatibility, oxygen supply, vascularization, and fibrotic responses. Additionally, it will cover the integration of these approaches with immunotherapy and gene editing. Regulatory hurdles for clinical application will also be discussed.
