Development of bioluminescent transdermal delivery system for wound healing

Light-based therapy has been widely investigated for various applications but their efficiency has been limited by the finite depth of light penetration. Bioluminescence as an internal light source has been considered a potential solution to overcome the penetration depth of light. This work introduces a bioluminescent transdermal delivery system using self-assembled polymersomes for wound healing. Luciferases are conjugated with carbon dots to emit near-infrared (NIR) light through bioluminescence resonance energy transfer (BRET) and induce wound healing by increasing collagen synthesis, promoting cell migration, and reducing early inflammation. Furthermore, the combination with rose bengal can increase crosslinking of collagen and other proteins, further enhancing the wound healing effect. Taken together, the polymersome-based bioluminescent system can offer effective transdermal delivery, improved stability, and prolonged therapeutic effects.
