Dual Targeted Polymersomes Impart Effective Delivery of Piperlongumine For Pancreatic Cancer Targeting

Natural drug regimens offer a marginal advantage over chemotherapeutic agents, particularly in terms of side effects. Therefore, we fabricated dual-targeting polymersomes (DTPS) loaded with a natural drug piperlongumine (PL), for effective pancreatic cancer treatment. A hyaluronic acid backbone was functionalized with DSPE-PEG-NH2, bis PEG amine, and phenylboronic acid to develop DTPS. The fabricated DTPS selectively recognized CD44/sialic acid overexpressed on pancreatic cancer cells, internalized, causing up to 80% cell growth inhibition, reducing the volume of 3D tumoroids with 58.3% of dead cells. The results demonstrated that PL-loaded DTPS were safe and effective for treating pancreatic cancer cells. Hence, the present study provides strong preliminary evidence that PL can be used as an alternative chemotherapeutic candidate with dual targeting polymersomes for human pancreatic cancer treatment.
