Reactive Polymer Based Near-Infrared Fluorescent Material Platform for Biomedical Applications

Fluorescent polymers with emission in the near-infrared (NIR) region are highly desirable imaging agents in biomedical applications; however, their synthesis remains challenging due to low reaction efficiency, long reaction time, and limited compatibility. In this study, we graft amine-functionalized IR775 NIR dyes to a reactive poly(pentafluorophenyl acrylate)(PPFPA) polymer to prepare NIR fluorescent polymer. The versatility of the polymer is demonstrated in two applications. In the first application, the IR775-grafted PPFPA is further modified with poly(ethylene glycol)(PEG) and biotin to form nanoparticles in aqueous solutions. We demonstrate the selective binding of the nanoparticles to target cells and the enhancement in the fluorescence signal. In the second application, the IR775-grafted PPFPA is modified with peptides and PEG and then inserted into lipid nanoparticles(LNPs). We demonstrate the increased binding affinity of the LNPs to target cells and utilize it to investigate the LNP delivery route.
