Proteins are critical components for organisms and mediate various biochemical reactions that occur within cells. The novel design of hybrid protein is essential for a successful tissue repair. Herein, hybrid protein was engineered using glutathione (an antioxidant enzyme) and gelatin (a structural protein). The glutathione-gelatin hybrid (GG) performed as an oxidative stress down-regulator. For ROS-damaged salivary gland repair, GG was fabricated into a cell carrier via photo-crosslinking, and adipose-derived mesenchymal stem cell (AdMSC) spheroids were loaded into the GG cell carrier (GGH). The GGH with AdMSC spheroids system was applied to an incurable reactive oxygen species (ROS)-damaged salivary gland mice model. After irradiation, excessive residual ROS were neutralized by GGH, which modulated the pro-inflammatory reaction and tissue fibrosis. Additionally, tissue repair was accelerated by AdMSC spheroids. Antioxidant hybrid protein system is a promising cell/drug carrier for incurable tissue repair.
