Microgravity (μg) in space causes numerous physiological changes in astronauts, such as loss of muscle mass and bone density, which are very limiting for space missions. To overcome the μg, research is being conducted on the International Space Station, but the accessibility of research is limited due to the high cost and transport constraints. Therefore, various systems are developed to simulate the μg on the ground (e.g., clinostat). Ground-based simulators are not a definitive validation method for modeling μg in space due to the influence of external factors, such as shear stress, making it challenging to distinguish the impact of μg alone. In this study, we developed a cell culture system using neutral buoyancy to minimize the limitations of conventional μg-simulated systems. We investigated the behavior of stem cells (e.g., stemness maintenance, attenuated osteogenic differentiation, etc.) to verify that the system can mimic the μg of space.
