Rapid Formation of Cell Sheet using Thin Leaf-Stacked Structured Film

The development of cell sheets has led to major advances in tissue engineering. Currently, the thermos-sensitive Poly(N-isopropylacrylamide)-based technique to prepare cell sheets is commonly used. However, the conventional method still has the limitation that it takes a long period to prepare a cell sheet. To overcome this hurdle, we developed a novel technique to form a cell sheet in a shorter time than the conventional method. We made an alginate film coupled with leaf-stacked structures (LSS). Subsequently, the side with the LSS layer was then coated with platinum (Pt), resulting in a thin-LSS film (TLSS). Following the complete dissolution of the alginate layer, cells were cultured on the Pt-coated LSS layer side for 3 hours, resulting in a cell sheet. In this study, the morphology of the TLSS, the time required to form the cell sheet, the structure of the cell sheet, and the interaction between cells in a cell sheet were investigated.
