Xenogeneic-free polymer for culturing human intestinal stem cells to regenerate intestinal disease

Basement membrane extract (BME), such as Matrigel presents a significant challenge due to its undefined and heterogeneous nature, creating barrier for the development of cell therapeutics. To overcome the limitations of traditional culture system, we develop xenogeneic-free dish for intestinal stem cells (XF-DISC) through polymer screening test of 32 kinds of surfaces deposited by initiated chemical vapor deposition (iCVD) process on the cell culture dishes. The XF-DISC enables rapid propagation and facilitates long-term culture of intestinal stem cells (ISCs). Additionally, cryo-banking of ISCs for over 3 years is affordable with XF-DISC. In vitro intestinal tissue model is confirmed on XF-DISC without discernible differences compared to Matrigel-coated surface. Furthermore, ISCs cultured on XF-DISC can engraft and regenerate intestinal epithelium in both EDTA-induced injury and DSS-induced colitis models by direct transplantation into the mouse colon.
