Synthesis and Characterization of Chitosan-TPP Nanoparticles for Oral Administration of DNA Vaccines

Currently, the only method for vaccinating fish at farms is through individual injections, which is inefficient in terms of time, labor, and cost. To address this, we aimed to develop an efficient oral drug delivery system for fish vaccines using chitosan, tripolyphosphate (TPP), and chondroitin sulfate (CS)-bile acid. We successfully encapsulated plasmid DNA vaccines by mixing chitosan, TPP, and plasmid DNA. The resulting chitosan-DNA nanoparticles were then coated with CS-bile acid. Zeta potential measurements showed that the initial chitosan-DNA nanoparticle surface had a zeta potential of +15.11, which changed to -26.87 after CS-bile acid coating. The nanoparticles ranged in size from 230 nm to 295 nm, and their spherical shape was confirmed by FE-SEM. These particles are highly stable, with a size of 100 to 200 nm, making them suitable for oral administration to fish. Finally, we administered the nanoparticles containing DNA vaccines orally to fish to study their effects.
