Glucosamine-mediated NK Cell Surface Engineering for Targeting Glioblastoma

Due to the unique tumor microenvironment and immune system, current immunotherapies for glioblastoma pose challenges. NK cell therapy could be applied since its remarkable inherent ability to eliminate cancer. To enhance anticancer efficacy for targeting glioblastoma, we developed an ex vivo NK cell coating strategy using Glucosamine-HA-PEG-Lipid, composed of 1) lipid anchor for membrane engineering via hydrophobic interaction, 2) PEG for blocking intracellular penetration, and 3) both glucosamine and hyaluronic acid (HA) for simultaneous targeting GLUT1 and CD44, overexpressed on glioblastoma. Our research demonstrated effective manipulation of NK cell surfaces facilitated targeting and eliminating glioblastoma in 3D tumoroid models without compromising intrinsic properties. Overall, our study suggests that biomaterial-based NK cell surface engineering could be a powerful tool in glioblastoma treatment.
