Extensive research has been conducted on the effects of molecular weight, polydispersity, and composition of poly(lactic-co-glycolic acid) (PLGA). However, the impact of monomer sequence distribution has been less studied, primarily due to the challenge of precisely controlling the monomer sequence in PLGA. In this study, we present a semibatch copolymerization strategy that allows for the production of sequence-controlled PLGA polymers by regulating the rate of comonomer addition. Using this method, we prepared a series of PEG-PLGA samples with comparable molecular weight and composition but varied sequence distributions. Additionally, PLGA-PEG-PLGA triblock copolymers with different sequence distributions were synthesized to investigate their effects. Our results demonstrate that altering the sequence distribution significantly influences physical properties, thermal properties, and drug release profiles.
