The development of in vitro transcribed mRNA (IVT mRNA)-based therapeutics and vaccines faces challenges due to the immunogenicity of IVT mRNA, which can trigger immune responses and reduce therapeutic efficacy. Traditional methods to mitigate this involve costly chemically modified nucleotides. An alternative approach, Additional Chimeric Element incorporated mRNA (ACE mRNA), combines conventional IVT mRNA with RNA/DNA chimeric elements, reducing immune responses and enhancing protein expression without the need for chemical modifications. This method restricts RIG-I and STING-mediated immune activation, offering a cost-effective and efficient solution for mRNA-based therapies/vaccines.
