Light-responsive gene delivery into a single living cell

On-demand delivery of genetic materials, such as plasmid DNA, siRNA, and CRISPR, into a desired location of single living cells is significantly important for precise cell manipulations and their therapeutic uses. However, rigid plasma membranes do not allow the negatively charged nucleic acids to be easily internalized into a target living cell. Here, we develop a mechanically robust and thin nanowire that enables delivery and release of pre-designed materials into a single living cell without both structural and physicochemical damages. Using chemical conjugation via photocleavable linkage, a polymeric nanowire was covered with high-density gene materials, followed by its injection into a target cell. When UV light was directly transmitted to the surface of the nanowire through the coupled optical fiber, the delivered genes were consequently released inside the living cell without optical damage, holding great potential for spatiotemporal gene delivery into diverse organelles (e.g. nucleus, nucleolus, and mitochondria).
