Preparation of an injectable in situ forming hydrogel combined with BMP2 for the osteogenic differentiation of hPLSCs

This study is the preparation of an injectable thermosensitive hydrogel, methoxy polyethylene glycol-(polycaprolactone-(N3)) block copolymer (PCL-N3), conjugated with covalent bone morphogenetic protein-2 (BMP2), targeting the osteogenic differentiation of human periodontal ligament stem cells (hPLSCs). O-propargyl-tyrosine (Opt) was site-specifically linked to BMP2 to produce BMP2-Opt with an alkyne group. The azide group of PCL-N3 and the alkyne group of BMP2-Opt are combined by a Cu(I)-catalyzed simple reaction. The PCL-BMP2 prepared by click reaction was injected into the subcutaneous fat with hPLSCs, and gel formation and osteogenic differentiation were confirmed by histological staining and gene expression analysis. In conclusion, we developed BMP2-Opt and an injectable PCL-BMP2 for osteogenic differentiation of hPLSCs. We expected that this injectable in situ forming PCL-BMP2 hydrogel could be used for non-invasive management of therapies for debilitating orthopedic conditions.
