Preparation and evaluation of an intratumorally injectable, click-cross-linked hyaluronic acid hydrogel

Primary method to treat cancer is a surgical method which can remove cancer tissues. A drug treatment method is used to remove remaining cancer tissues and prevent cancer metastasis by remaining cancer tissues. Doxorubicin (DRX) has excellent anticancer effects, but it is very toxic and has a short half-life in human body. That is why release of the drug must be well controlled. To overcome side effects of DRX and enhance efficacy of DRX, we prepared click-cross linkable hyaluronic acid hydrogel that can be directly injected into cancer. We individually introduced trans-cyclooctene (Tc) and tetrazine (Tz) into hyaluronic acid (HA) so that the Tc-modified HA and Tz-modified HA immediately formed DRX-containing hydrogel after being injected and DRX was sustainably released. Cross-linked HA hydrogel consistently inhibited tumor growth. In conclusion, we prepared a hydrogel capable of forming a drug depot within tumor and the anti-cancer effect of DRX loaded hydrogel increased.
