pH-Responsive Polymers Based on Cyclic Acid Amide for Controlled Drug Release

Our body tissues and intracellular organelles show distinct pH ranges. Especially, inflammatory tissues and cancer tissues exhibit weakly acidic pH than that of normal tissues. This mildly acidic environment can serve as a trigger for a selective drug delivery so many pH-responsive polymers that can change their chemical structure and properties in response to pH change have been developed and utilized in the biomedical fields. Herein, we report a new type of pH-responsive polymer containing cyclohexane 1,2-dicarboxylic acid anhydride monomers synthesized via RAFT polymerization. The polymers with such anhydride can readily react with amines to form β-carboxylic acid amide bonds. This acid amide bond can degrade and release amine in response to pH change due to its sterically fixed α, β-bond. At acidic pH, they degraded and released amine, while bonds are quite stable under neutral or basic pH. These polymers can be applied to develop materials with pH-responsive for the drug delivery system.
