Fabrication of 3D Adipose Tissue Using Engineered 3T3-L1 Spheroids

3D cell spheroids with biomaterials have been studied for their ability to control cell function through direct signal delivery. Recent research focuses on 3D artificial adipose tissue for reconstruction and drug screening, but fabrication has been limited. Here, we applied an engineered nanofibers to transmit internal adipogenic signals to 3T3-L1 within spheroids. We fabricated adipogenic nanofibers (AF) via polydopamine assisted insulin immobilization on indomethacin-incorporated nanofibers (IF) and introduced AF to form a spheroid with 3T3-L1 cells. Then, we quantified the concentration of indomethacin in the fibers and characterized the surface chemistry, immobilization efficiency, and release kinetics of insulin. Also, we investigated the proliferation and adipogenic differentiation of AF incorporated 3T3-L1 spheroids. Further, we encapsulated AF spheroids in Gel-MA hydrogel to create 3D adipose tissue construct and investigated the cellular behavior of 3T3-L1 cells in hydrogels.
