H<sub>2</sub>O<sub>2</sub>-Activatable ursodeoxycholic acid polymeric nanoparticle for the treatment for fatty liver diseases

Non-alcoholic fatty liver disease(NAFLD) has become the most common chronic liver disease, causing a tremendous clinical burden. Despite the increasing prevalence of NAFLD, definitive medical treatment has not been established. Here, we propose a new therapeutic approach for NAFLD exploiting oxalate linkages that can be cleaved by H2O2 overproduced during NAFLD. We synthesized poly(ursodeoxycholic acid oxalate) (PUOX) with H2O2-scavenging peroxalate ester bonds as a polymer prodrug of ursodeoxycholic acid (UDCA). PUOX could be formulated into nanoparticles with 200nm~. In the cell culture study and mouse models of NAFLD show antioxidant and anti-inflammatory effects by releasing UDCA in the region of fatty liver in a H2O2-triggered manner. The nanotherapeutics of PUOX in polymeric systems can be a promising approach to improve the efficacy for various H2O2-associate diseases.
