Adhesive composite hydrogel patch based on a polyaspartamide derivative to treat atopic dermatitis

A polyaspartamide derivative, poly(N-2,3-dihydroxypropyl aspartamide) (PDHPA) is a sort of poly(amino acid) and is comprised of a peptide backbone and 1,2-dihydroxy functional groups. With using boric acid (BA) as a crosslinker, a hydrogel based on reversible bond between polymer and crosslinker can be prepared. PDHPA-BA hydrogel shows dynamic reversibility, self-healing, and pH-triggered sol-gel transition. By introducing mesoporous silica nanoparticles (MSNs) with abundant hydroxyl groups, several features including mechanical performance, adhesion performance, and self-healing rate of the PDHPA-BA hydrogel are improved. The high surface area and mesopores of MSNs make it possible to load hydrophobic drugs such as dexamethasone on the hydrogel. Owing to the innate adhesiveness of the hydrogel and appropriate assistance of MSNs, PDHPA-BA/MSN hydrogel could be successfully applied as a transdermal drug delivery patch to treat atopic dermatitis using mouse model.
