Nano-analgesic agents with HA-PLGA as treatment of Chemotherapy Induced Peripheral Neuropathy (CIPN).

Cancer patients treated chemotherapy with neurotoxic anticancer drugs can develop side effects, particularly chemotherapy-induced peripheral neuropathy (CIPN), which is often painful and can lead to premature chemotherapy discontinuation. As analgesic agents for the treatment of CIPN, Aconite roots (AR) have been widely investigated, but AR contains toxic aconitine. In this study, we developed nano-analgesic agents with HA-PLGA to delivery non-toxic aconine from toxic aconitine by heating. Aconite root extract was encapsulated in HA-PLGA nanoparticles through W/O/W emulsion method. The appropriate heating time was 48 hours and successful extraction was characterized by HPLC. In addition, the spherical HA-PLGA/AR extract with a size of 200nm was confirmed by TEM and DLS. The analgesic property was confirmed by in vitro release and neurite outgrowth test using PC-12 cells. In conclusion, these results indicated this nanoplatform can be extensively exploited in not only CIPN treatment but various biomedical applications.
