Highly robust multilamellar lipid nanovesicles were prepared through the intervesicular self-assembly of pre-formed cationic 1,2-Dioleoyl-3-trimethylammonium-propane (DOTAP) large unilamellar vesicles (LUVs) with hydrolyzed collagen peptides (HCPs). The LUVs undergo vesicle-to-vesicle assembly and structural reorganization through polyionic complexation with HCPs, resulting in the formation of multilamellar collagen-lipid vesicles (MCLVs). HCP acted as a non-covalent cross-linker for lipid LUVs to induce their self-assembly into MCLVs, which have excellent structural stability against pH, ionic strength, and surfactants. In addition, the structural stability of MCLVs remains unchanged against repeated freeze-thaw stresses, demonstrating the excellent stabilization effect of the HCPs on the MCLVs. This work allows an appealing way for fabricating simple and robust MCLVs without covalent cross-linkers, organic solvents, and specialized instruments.
