Adipocyte death induced by gas-generating polymer nanoparticles

Surface-modified polymer nanoparticles are being currently used to improve treatment efficacy and reduce side effects of various diseases, including obesity. Poly(lactic-co-glycolic acid) nanoparticles loaded with calcium carbonate mineral were prepared as gas-generating nanoparticles under acidic endosomal conditions. The surface of the gas-generating nanoparticles was modified with functional peptides to improve the intracellular delivery. Gas-generating polymer nanoparticles showed an significant adipocytolitic effect in vitro and in vivo. The reduction of adipocyte number was mainly caused by membrane disruption after intracellular gas-generation. The process of cell death after membrane disruption was investigated in vitro. Conclusively, surface-functionalized gas-generating nanoparticles were useful as a targeted delivery vehicle to adipocytes, and carbon dioxide gas released from the nanoparticles useful to reduce the adipocytes by membrane disruption.
