Overcoming “PEG dilemma” in amphiphilic PEG-b-PCL using the alternative multifunctional polyglycerol (PG) for the hydrophilic block and the modifiable crystalline poly(caprolactone-co-lactide) (PCLL) for the hydrophobic block.

Despite advantages of PEG-b-PCL on circulation for chemotherapy, limited modification site on PEG block and high crystallinity of PCL block initiate difficulties in cellular targeting and colloidal stability. In my works, hydrophilic group was replaced by PG for enhancing ligand incorporation efficiency, and hydrophobic group was modified with the addition of L-lactide to create adjustable crystallinity. In this presentation, I report three correlated studies on PG-b-PCL, PEG-b-PCLL, and PG-b-PCLL. The research results showed that folate can incorporate into PG up to 21%, and the tumors were suppressed by 4 times lower dose than clinical one. Reducing the crystallinity level in hydrophobic blocks indeed improved the stability of the nanoemulsions. Overall, the polymers are promising candidate to completely substitute and suppress the dependence on PEGylated nanocarriers in cancer chemotherapy.
