Self-Immolative Poly(benzyl ether)-Based Amphiphiles: Synthesis and Demicellization via Head-to-Tail Depolymerization

This presentation describes the design of degradable polymer amphiphiles capable of triggered demicellization via head-to-tail depolymerization. The amphiphiles could be synthesized from two quinone methide monomers through anionic polymerization and form micelles under aqueous conditions. In particular, selective depolymerization behavior of the copolymers enabled triggered demicellization that can proceed completely or partially under the control of applied signals. The designed micelles were further demonstrated to be stably loaded with hydrophobic drugs and completely release them on demand as being depolymerized. The depolymerization products were further confirmed to be biocompatible via cell viability tests
