Rapid detection of COVID-19 in human nasopharyngeal swap specimen with polymer-assisted nucleic acid extraction and digital PCR

Coronavirus disease 2019 (COVID-19) diagnostics have significantly prevented early-stage transmission of COVID-19 viruses. Nasopharyngeal swabs are widely used for the COVID-19 virus yet require multiple purification steps to reduce false positives or false negatives. To simplify the RNA extraction from the patients’ clinical samples, we modified the surface of the substrates with a tertiary aminated polymer as a thin film form. The polymer we adopted is poly(dimethyl aminomethyl styrene) (pDMAMS) synthesized via initiated chemical vapor deposition. By simply transferring the clinical samples to the tubes, the RNAs could be captured on a positively charged pDMAMS layer. From the surface analysis and PCR, the successful capture of RNA was confirmed, with a capture efficiency of 100 %. Finally, the performance of diagnosing COVID-19 with our platform was tested with patients’ clinical samples. Sensitivity and specificity turned out to be 90 % and 100 %, within 25 minutes of pre-treatment.
