Design of Copolymer Inhibiting Protein Denaturation during Cryopreservation of the Protein

Cyropreservation of protein is important for long term storage. Also it results in development of protein processing and biopharmaceutics. However freezing can induce instability of protein because salt concentration increase nonequally. As well, recrystallization occurs in thawing, forming additional interfaces where protein may absorb and denature. Here in, we design polymer mimics of AFGP. AFGP is composed with threonine/alanine backbone and sugars that bind to secondary alcohols of threonine. As inspired by this structure, we synthesize copolymers whose backbone consist of threonine/alanine in varying ratios and are functionalized with succinate intead of sugar in differrent prportions. We expect for backbone to be able to bind with the ice interfaces, inhibiting recrystallization and for succinate to act like the sugar that inhibit ice formation by interacting with water. It can be confirm through IRI and INI assay. Among our four polymers, P3 proved to preserve the protein activity better than PVA having good IRI activity then be an excellent cyroprotectant.
