Biodegradable Albumin-based Nanocarriers for Intracellular Drug Delivery

Nanosized particles have been extensively studied for intracellular delivery of therapeutic agents. Recently, bovine serum albumin (BSA) has gained increasing attention as a biomaterial for nano-particulate drug delivery system because of its good biocompatibility, high stability, and facile surface functionalization. Here, we report a cationic BSA-based nanocarrier for enhanced intracellular delivery of therapeutic agents to cancer cells. After preparing uniform BSA nanoparticles through simple desolvation and crosslinking steps, cationic poly-L-lysine (PLL) was coated on the surface of BSA nanoparticles to generate surface features favorable for cell permeation. The properties of BSA nanoparticles as a drug delivery carrier were assessed in terms of loading capacity, encapsulation efficiency, and intracellular delivery efficiency. The cationic BSA nanocarriers showed high loading efficiency and improved cell permeation performance compared to non-functionalized BSA nanoparticles.
