Novel Non-cytotoxic Fluorescent Skeletons based on Quercetin Derivatives

Many fluorescent molecules are used in various cell imaging. However, the reported organic fluorescent probe showed limitations for further applications due to cellular toxicity in high concentrations. Therefore, it is important to discover a non-toxic fluorescent probe. Here, we reported a novel molecular skeleton for fluorescence dyes using biocompatible flavonoids. Quercetin (QT) is a representative bioflavonoid and has shown many applications in metal sensors, anticancer activity, and anti-inflammatory drugs. We synthesized the quercetin derivative through the etherification reaction between the phenol group of quercetin and the oligoether chain. Interestingly, penta-substituted QT showed stronger fluorescence intensity compared to tetra-substituted QT. The fluorescent phenomenon was investigated based on solvatochromism, dye aggregation, and DFT calculations. Furthermore, the MTT analysis showed that the fluorescent quercetin derivative is non-toxic as a new biocompatible fluorescent candidate.
