Coordinative Double Hydrophilic Polyether Micelle for Controlled Delivery of Cisplatin

While its widespread use in the treatment of numerous cancers, cisplatin suffers from several drawbacks such as high toxicity and low selectivity, which necessitates the development of drug delivery system. Herein, we introduce a double hydrophilic block copolyether of poly(ethylene oxide)-b-poly(glycidoxy acetic acid) (PEO-b-PGA) synthesized via anionic ring-opening polymerization using a novel oxazoline-based epoxide monomer, followed by a subsequent acidic deprotection. The successful synthesis of the polymers was characterized by a series of 1H, 13C-NMR, and GPC spectroscopies. The coordinative interaction between cisplatin and carboxylate group in PGA block induced the formation of the micelles from the PEO-b-PGA. The loading efficiency of the micelles and the controlled release of cisplatin were carefully characterized. Considering the micelles' high biocompatibility and pH-responsive characteristics, the polymeric micelle will provide a new strategy for the controlled delivery of cisplatin.
