Characterization of a basic peptide-PAMAM G2 conjugate dendrimer as a gene carrier

In this study, the polyamidoamine dendrimer generation 2 (PAMAM G2) was conjugated with a basic peptide composed of arginine and histidine amino acids and was characterized as a gene carrier by investigating its DNA binding ability, cytotoxicity, and transfection efficiency. The multivalent basic peptides of the conjugate dendrimer can induce the electrostatic interaction between the conjugate dendrimer and plasmid DNA, and can increase the proton sponge effect of the polymer which causes the endosomal escape of polyplexes during endocytosis. The overall synthesis yield of the peptide-PAMAM conjugate dendrimer was confirmed to be over 92%. DNA binding ability was verified through agarose gel electrophoresis. Cytotoxicity and transfection efficiency was confirmed using A549 lung cancer cells. The conjugate dendrimer showed low toxicity and potent transfection efficiency at 12 and 16 polymer/DNA weight ratios.
