Hyaluronic acid based hydrogel filler encapsulating adipose derived stem cell extracellular vesicles reprograms the dermis microenvironment

As the social atmosphere that wants to look healthy and young grows with the aging of the population, the filler market, a representative of the anti-aging industry, has been under rapid development. In the process of aging, the dermal structure is impaired due to a decrease in fibroblast and dysregulation of the extracellular matrix(ECM), so a dermal filler has emerged to repair it. Here, a high molecular weight hyaluronic acid(HMWHA)-based hydrogel filler loading the adipose derived stem cell extracellular vesicles(ASEVs) was manufactured to reprogram dermis into anti-inflammatory environment. Hydrogel was formed by crosslinking 1000kDa of HA with 1,4-butanediol diglycidyl ether(BDDE), and ASEVs loading hydrogel filler was finally prepared through simple mixing with ASEVs. This filler system showed the potential as anti-aging filler that improves the dermal microenvironment by enhancing the expression of CD301b on macrophage associated with collagen production by the action of ASEVs.
