Ultrasound-responsive gold-crosslinked hyaluronic acid depot as a potential carrier for on-demand cargo release in cancer therapy

Temporal and quantitative manipulation of the cargo release has always been a challenge in cancer therapy due to the cancer treatment minimum effective concentration. Therefore, we designed an ultrasound-triggered hyaluronic acid (HA) hydrogel depot with gold-clustered polymeric nanoparticle crosslinked to the dopamine-functionalized HA (dHA) to release cargo on-demand. This hydrogel exhibited great retention in the structure, low cargo leakage and superior biostability for about 4 weeks due to the presence of cross-linking between gold clusters and dHA. The reason is that the gold clusters could be dissociated upon ultrasound (US) irradiation, which lead to an efficient cargo release and cancer cell death. Moreover, spiky structure of gold surface can provide an excellent sonothermal property that can elevate heat up to 53.7oC upon US irradiation. These results highlight the significant potential of this injectable hydrogel for future in vivo applications in tumor therapy.
