Pneumatic valve-based microfluidic device for the detection of hEPO gene

Since erythropoietin (EPO)- and EPO-related formulations have been widely used for the endurance of athletes, EPO gene has been identified as a main target of abuse in gene doping. Therefore, microfluidic devices were used to develop systems that detect them. The newly developed gene doping chip is a pneumatic valve system and has a loop reactor that can be incubated through circulation in the channel. In addition to the well-known advantages of reduced sample volume and short analysis time, these devices also offer significant advantages in other ways. For example, low-mass samples can be easily handled, and fluid flow can be controlled quickly and accurately using additional solenoid valve functions. For immunoprecipitation, the shape of the loop reactor in the channel was optimized, and demonstrations using color dyes and magnetic beads were also possible within the optimized loop. We will finally create a CRISPR system-based sensing platform using this gene doping chip. Thus, gene doping chips may be considered a suitable method for target detection in gene doping.
