Gelatin-based Bioactive Hydrogels Delivering Oxygen and Calcium Ions for Enhanced Angiogenic Effect and Osteoblast Proliferation

Recently, polymeric hydrogels have been widely used as a therapeutic carrier for delivering cells or bioactive factors to improve vascularized bone tissue regeneration. Among these, growing evidence demonstrates that oxygen (O2) and calcium ions (Ca2+) play a critical role in bone regeneration, such as cell proliferation and angiogenesis. Herein, we report gelatin-based bioactive hydrogels delivering O2 and Ca2+ for vascularized bone tissue regeneration. This hydrogel is formed by a calcium peroxide-mediated crosslinking reaction. Also, our hydrogel has a controllable mechanical property and release kinetics of O2 and Ca2+ depending on the composition in vitro. Notably, our hydrogel enhances pre-osteoblast proliferation and upregulates the expression of vascular endothelial growth factors from mesenchymal stem cells. In sum, our O2- and Ca2+-releasing hydrogel has the potential as a bioactive matrix for vascularized bone tissue regeneration.
