Development of Versatile Antifouling Surface Coating by Mimicking Melanogenesis

Inspired by the melanogenesis found in nature, we report tyrosinase-mediated antifouling surface coating by synthesizing a tyrosine-conjugated sulfobetaine derivative (Tyr-SB). Our Tyr-SB contains zwitterionic sulfobetaine group to express antifouling capability and tyrosine whose phenolic amine group acts as a dormant coating precursor. In contrast to catechol amine derivatives which are usually prone to auto-oxidation under ambient air conditions, our phenol amine based, Tyr-SB is stable against auto-oxidation and are enzymatically oxidized only in the presence of tyrosinase to initiate melanin-like oxidation. When surface of interest was introduced during the oxidation process, poly(Tyr-SB) film was formed, and thereby showed superhydrophilic property. The poly(Tyr-SB) coated surface showed significant decrease in protein and cell fouling. Because our developed coating strategy can be applied toward various surfaces under mild aqueous conditions, we expected that it can be a useful tool to achieve various biomedical applications, including biosensors and medical devices.
