Spheroids-laden hydrogel with spatially confined delivery of signaling molecules for engineering 3D complex tissue

Engineering osteochondral tissue by using spheroids and hydrogels, consisting of two layers of bone and cartilage tissue, may be a potential method. However, the issues such as heterogeneous distribution of spheroids, limited sprouting of cells, inefficient delivery of inductive factors, and delamination of layers should be addressed. In this study, we developed the spheroid-laden bilayer hydrogel with controlled mechanical strength, in which effective cell sprouting and homogeneous spheroid distribution were achieved. In particular, we prepared the composite spheroids incorporating poly-i-lactic acid nanofibers coated with osteo- or chondro-inductive growth factors. The adipose-derived stem cells were differentiated into osteoblasts and chondrocytes in each layer, respectively, which would be a platform to address delamination and limited regeneration problems.
