Fabrication of Reactive Powder-type Hemostatic Agents Containing Blood Coagulation Factors Made of Dialdehyde Starch and Hyaluronic Acid

Persistent and severe bleeding can lead to death. To accelerate hemostasis for narrow areas, research on powder-type hemostatic agents has been actively reported. It needs to improve tissue adhesion and blood clotting ability to be used efficiently without disturbing the wound area. In this study, the hydroxyl group of starch was oxidized to dialdehyde groups, and hyaluronic acid was crosslinked through glyoxal to enhance tissue adhesive force. Ca2+ ions and thrombin are incorporated into crosslinked powders as blood coagulants. The hemostatic ability was controlled by the composition of DAS/HA, the amount of crosslinking agent, and particle size. As a result, DAS/HA/CaCl2/thrombin powder showed a better coagulation property than the control at in vitro. In vivo study with a rat hepatic hemorrhage model, fabricated powder showed less bleeding time and bleeded amount compared to control. DAS/HA powder introduced with Ca2+ ions and thrombin is expected to be widely applicable to surgical operations.
