Enhanced Cancer Vaccination via Nanomicelle-Formed by Dissolving Microneedles

Compared to subcutaneous injection, microneedle (MN) has been considered as a minimally invasive skin drug delivery method that can overcome skin problems. Among them, dissolving microneedles (DMN) are attracting a lot of attention due to their simple fabrication and easy to apply.
DMN dissolves rapidly upon contacted with intradermal fluids, enabling efficient drug delivery and minimizing the risk of inflammation. However, since DMN is formulated from hydrophilic polymers and dissolve rapidly in intradermal fluids after administration, it is often difficult to apply as a water-insoluble components.
In this study, we developed amphiphilic polymer-based DMN and used them as carriers for transdermal drug delivery system of hydrophobic adjuvant (R848) and hydrophilic cancer model antigen (OVA). Application of MN containing R848 and OVA to skin cancer mice of EG7-OVA induced significant levels of antigen-specific humoral and cellular immunity, resulting in significant anti-cancer activity.
