Directly and site-selectively modifying a native protein with electrophile ligand-tagged nucleic acid biopolymers

Due to frequent requirements of protein functionalization, many protein labeling studies have drawn attention in biotechnology. However, direct modifications of native proteins have limitations on maintaining their inherent activities; for proper protein folding, the chemical modifications should not be indiscriminate, but be conducted under mild conditions. To address them, we create a novel strategy enabling a direct and site-specific protein modification by electrophile ligand-tagged nucleic acid biopolymers. Without impairing protein activities, our engineered DNA aptamers specifically recognize target proteins, and the delivered electrophile ligands are subsequently transferred to the desired nucleophilic residues on the protein surface. The ligands contain an alkyne group that can react further with an azido group; the availability of the following bioorthogonal click reactions holds great potential for our strategy to be widely applicable for the elaborate generation of various protein conjugates.
