Effects of stiffness of collagen-coated and hydrogel substrates on cell-cell interaction

This study investigates the effect on fibroblasts by stiffness and interactions with cells. We cultured fibroblasts on the non-coated, collagen-coated, and collagen hydrogel substrates. Also, fibroblast spheroids were cultured on each substrate and compared to mono-layered cells. The mechanical property of cellular substrates is known to affect the cell behaviors such as proliferation, migration, and differentiation. We performed an MTT assay and quantification for collagen and glycosaminoglycan. In addition, AFM was conducted to identify the roughness of the collagen surfaces. Moreover, we assessed the expression of YAP and E-cadherin through immunocytochemistry. In conclusion, a more useful way is the spheroid culture on the soft collagen matrix because it is closer to the real cellular environment than the conventional 2D cell culture.
