Development of multi-functional extracellular matrix (ECM)-mimetic 3D scaffold for vascularized bone tissue regeneration

Functionalized scaffolds have been widely used for bone tissue engineering. However, low biocompatibility, limited host integration, difficulty in delivery of multiple signaling molecules remained problems. In this study, we developed biomimetic scaffolds enabling delivery of various instructive cues by combining decellularization of multi-cellular spheroids. We prepared a 3D printed scaffold filled with human adipose-derived stem cell (hADSC) spheroids (MS) incorporating osteoinductive mineral-coated nanofibers, decellularized with Triton-X (dC-MS). We found that osteoinductive and pro-angiogenic biomolecules were preserved after decellularization. The in vivo implantation of dC-MS onto a mouse calvarial defects demonstrated complete host tissue integration and enhanced vascularized bone tissue regeneration. In conclusion, our multi-functional scaffold could be potentially used for the regeneration of complex bone structure.
